چکیده
The crucial role of the myofibroblast in wound healing and fibrosis development is well established. This review discusses the mechanisms of myofibroblast action and the new findings that may develop into therapeutic strategies during the next few years.

Mechanoregulation of the Myofibroblast in Wound Contraction, Scarring, and Fibrosis: Opportunities for New Therapeutic Intervention.
SIGNIFICANCE Myofibroblasts are responsible for wound closure that occurs in healed acute wounds. However, their actions can result in disfiguring scar contractures, compromised organ function, and a tumor promoting stroma. Understanding the mechanisms regulating their contractile machinery, gene expression, and lifespan is essential to develop new therapies to control their function. RECENT ...
متن کاملHoming in on the hepatic scar: recent advances in cell-specific targeting of liver fibrosis
Despite the high prevalence of liver disease globally, there are currently no approved anti-fibrotic therapies to treat patients with liver fibrosis. A major goal in anti-fibrotic therapy is the development of drug delivery systems that allow direct targeting of the major pro-scarring cell populations within the liver (hepatic myofibroblasts) whilst not perturbing the homeostatic functions of o...
متن کاملTowards an anti-fibrotic therapy for scleroderma: targeting myofibroblast differentiation and recruitment
BACKGROUND In response to normal tissue injury, fibroblasts migrate into the wound where they synthesize and remodel new extracellular matrix. The fibroblast responsible for this process is called the myofibroblast, which expresses the highly contractile protein alpha-smooth muscle actin (alpha-SMA). In normal tissue repair, the myofibroblast disappears. Conversely, abnormal myofibroblast persi...
متن کاملIdiopathic pulmonary fibrosis: a disease with similarities and links to cancer biology.
Several clinical trials have recently targeted specific pathways implicated in the pathogenesis of idiopathic pulmonary fibrosis (IPF). However, IPF remains plagued by a median survival of 3 yrs and emphasises the need for further research with new insights and perspectives. The prevailing pathogenic hypotheses assume that either an inflammatory process or an independent epithelial/fibroblastic...
متن کاملNew insights into the prevention and treatment strategies for hypertrophic scars and keloids
Hypertrophic scars and keloids are fibrosis abnormalities associated with the accumulation of collagen and extra cellular matrix components. These scars are caused by abnormal wound healing, which may occur after skin injuries caused by surgery, trauma, burns, etc. and may have a large impact on the patients’ quality of life. Hypertrophic scars and colloids in addition to aesthetic problems can...
